Overview This in vitro transcribed firefly luciferase mRNA incorporates Cy3-labeled nucleotides, enabling direct visualization of mRNA delivery and cellular uptake by fluorescence microscopy using a Cy3 filter set. Quantitative analysis of translation efficiency can be performed through a luciferase reporter assay.

  • 5' Cap Structure: Available with ARCA (Anti-Reverse Cap Analog; m7(3'-O-methyl)-G(5')ppp(5')G, Cap 0) or CleanCap AG (m7G(5')ppp(5')(2'-O-methyl-A)pG, Cap 1) to enhance mRNA stability and translational efficiency.
  • Nucleotide Modifications: Incorporates Cy3-UTP and N1-methyl-pseudouridine-5'-triphosphate at a 1:3 molar ratio, as well as 5-methylcytidine triphosphate (5mCTP), to improve mRNA stability and reduce innate immune activation.
  • Poly(A) Tail: 180 nucleotides for optimal translation.
