Description DNA Polymerase I, a mesophilic enzyme derived from the polA gene of Escherichia coli expressed recombinantly in Escherichia coli, catalyzes 5’ to 3’ DNA synthesis with 3’ to 5’ and 5’ to 3’ exonuclease activities, enabling nick translation during DNA synthesis.
Applications
  • Nick translation of DNA with DNase I to generate high-specific-activity probes
  • Second-strand cDNA synthesis with RNase H

Unit Definition One unit is defined as the amount of enzyme required to incorporate 10 nmol of dNTPs into acid-insoluble material in 30 minutes at 37°C under standard DNA polymerase assay conditions.
