| Background | Protocadherin FAT1 is a large single-pass transmembrane protein whose extracellular domain comprises 34 cadherin repeats, 5 EGF-like domains, and a laminin-G-like domain. The intracellular domain contains a nuclear localization signal and regulates gene expression upon activation. FAT1 modulates Hippo, TGF-β, and Wnt/β-catenin signaling pathways in colorectal and other cancers, promoting cell polarization and migration. FAT1 is expressed in epithelial, endothelial, and smooth muscle cells, where it regulates cell-cell interactions and responses to injury. Soluble FAT1 may serve as a potential biomarker for cancer diagnostics. |
