Design a matched parent-and-variant campaign for solubility, expression, stability, binding, or function. The workflow combines sequence liabilities and solubility/aggregation analysis; constrained proposals from protein language models, ProteinMPNN-family inverse folding, and rational design; structure/interface checks; and, when justified, OpenMM ensemble analysis. Antibody projects add CDR, Vernier-zone, and VH/VL-interface protection plus a method-separated humanization matrix using operational Sapiens, Hu-mAb-style, HuMatch, germline/template, and structural-consensus evidence. OASis, AbNatiV, HuDiff, CUMAb, and related methods are included only when their database, implementation, and commercial-use gates are satisfied. Deliverables include exact parent and variant FASTA, mutation maps, structures, per-method scores, provenance, and a wet-lab test panel.
