Design or optimize VHH candidates from a scaffold and antigen or epitope constraints. The sequence-first workflow performs VHH numbering and CDR mapping, framework-preserving proposal, physicochemical and liability triage, diversity-aware ranking, and fail-closed structure or complex assessment with available NanoBodyBuilder2-, ESMFold-, AlphaFold-Multimer-, Boltz-, RFantibody-, or ProteinMPNN-family adapters. Deliverables include exact FASTA sequences, mutation and CDR maps, ranked method-specific evidence, structures when supported, and complete lineage/provenance. Nativeness tools with academic or model-weight restrictions are used only after authorization. Computational ranking is not evidence of affinity, specificity, expression, or stability.
